EMMPRIN in extracellular vesicles from peritoneal mesothelial cells stimulates the invasion activity of diffuse-type gastric cancer cells

چکیده
Peritoneal metastasis of gastric cancer (GC) results in extremely poor prognoses. The peritoneal cavity is covered by a monolayer mesothelial cells (PMCs). Interactions between GC and PMCs might play pivotal role metastasis. Extracellular vesicles (EVs) correlate with intercellular communication. Although communication be associated the metastatic process, EVs from remains unclear. We investigated effects on cells. Three cell lines (OCUM-12, NUGC-3, MKN74) four were used. derived invasion migration evaluated Matrigel assay. Factors contained PMC analyzed; extra-cellular matrix metalloproteinase inducer (EMMPRIN) was detected EVs. an EMMPRIN inhibitor invasion-stimulating activity examined. expressions 110 GCs immunohistochemistry. significantly promoted diffuse-type cells, i.e., OCUM-12 NUGC-3 stimulated inhibited inhibitor. A high expression worse cancer-specific survival peritoneal-recurrence-free survival. stimulate malignant progression GC. useful prognostic marker recurrence patients.

MiR-493 suppresses the proliferation and invasion of gastric cancer cells by targeting RhoC
Objective(s):MiRNAs have been proposed to be key regulators of tumorigenesis, progression and metastasis. However, their effect and prognostic value in gastric cancer is still poorly known. Materials and Methods: Gastric cancer cell lines were cultured. Tissue samples obtained from 36 gastric cancer patients were used for quantitative real-time PCR (qRT-PCR) analysis. The tissue microarrays (T...
متن کاملThe role of human peritoneal mesothelial cells in the fibrosis and progression of gastric cancer
Peritoneal dissemination is the most frequent metastatic pattern of scirrhous gastric cancer. However, despite extensive research effort, disease outcomes have not improved sufficiently. Tumor progression and metastasis result from interactions between cancer and various cells in the stroma, including endothelial cells, immune cells and fibroblasts. Fibroblasts have been particularly well studi...
متن کاملLysophosphatidic acid is constitutively produced by human peritoneal mesothelial cells and enhances adhesion, migration, and invasion of ovarian cancer cells.
Lysophosphatidic acid (LPA) is both a potential marker and a therapeutic target for ovarian cancer. It is critical to identify the sources of elevated LPA levels in ascites and blood of patients with ovarian cancer. We show here that human peritoneal mesothelial cells constitutively produce LPA, which accounts for a significant portion of the chemotactic activity of the conditioned medium from ...
متن کاملTransduction of soluble Flt-1 gene to peritoneal mesothelial cells can effectively suppress peritoneal metastasis of gastric cancer.
The prognosis of gastric cancer with peritoneal metastasis has not improved. Despite many promising studies, gene therapy has limited clinical application because of the lack of suitable vector systems to enable selective gene transduction to tumor cells. The aim of this study was to clarify whether gene therapy targeted to peritoneal mesothelial cells (PMCs) can inhibit peritoneal disseminatio...
متن کاملPeritoneal repairing cells: a type of bone marrow derived progenitor cells involved in mesothelial regeneration
The peritoneal mesothelium exhibits a high regenerative ability. Peritoneal regeneration is concomitant with the appearance, in the coelomic cavity, of a free-floating population of cells whose origin and functions are still under discussion. We have isolated and characterized this cell population and we have studied the process of mesothelial regeneration through flow cytometry and confocal mi...
